吡咯烷二硫代氨基甲酸盐抑制胆固醇诱导的人肝细胞C反应蛋白的表达

【制中药论文】作者:唐俊利,徐海燕,刘喜平,钱民章【摘要】 目的 研究核因子κB(NF-κB)信号通路阻断剂吡咯烷二硫代氨基甲酸盐(PDTC)对胆固醇诱导的人肝细胞C反应蛋白(C-reactive protein, CRP)表达的影响。方法 培养肝细胞L-02并将其分为3个组,即胆固醇作用组、PDTC+胆固醇作用组和对照组,分别作用细胞48 h。采用Western blot和免疫比浊法分别检测细胞内和细胞培养液中CRP含量,比较有无PDTC作用下CRP表达量的变化。结果 胆固醇能显著诱导人肝细胞C反应蛋白的表达(P<0.05),且其诱导效应随浓度增加而增强;加入PDTC阻断NF-κB信号通路后,胆固醇诱导人肝细胞CRP基因表达的作用减弱(P<0.05)。结论 PDTC阻断NF-κB信号通路后,CRP表达量下降,提示NF-κB信号通路可能参与了胆固醇诱导CRP基因表达的过程。 【关键词】 C反应蛋白;胆固醇;动脉粥样硬化;核因子κB ABSTRACT: Objective To study the effects of PDTC on cholesterol induced C-reactive protein gene expression in L-02 cell line.
